 摘要:目的   探讨高脂、高糖饮食诱导的大鼠胰岛素抵抗对血管平滑肌细胞大电导钙激活钾通道(BKCa)功能的影响及其机制。方法   采用膜片钳技术检测胸主动脉和肠系膜动脉平滑肌细胞BKCa的功能;分别用免疫印记法和实时定量PCR法测定BKCa亚单位(α和β1亚单位)的蛋白和mRNA表达水平。结果   与对照组比较,胰岛素抵抗组大鼠胸主动脉和肠系膜动脉平滑肌细胞宏观电流密度均减低,100 nmol/L iberiotoxin使胰岛素抵抗组细胞在+60 mV时的最大电流幅度较对照组显著降低(P<0.01)。β1亚单位蛋白和mRNA表达在胰岛素抵抗组较对照组明显减低(P<0.05)。结论   胰岛素抵抗时无论大血管还是微血管BKCa功能的下降与β1亚单位表达下调有关。
Abstract:
 ABSTRACT: Objective   To investigate the function of large conductance Ca2+-activated K+ channel (BKCa) in vascular smooth muscle cells of rats with insulin resistance (IR) induced by a high-fat and high-sugar diet. Methods   The present study investigated the BKCa current and subunit expressions in smooth muscle cells from aortas and mesenteric arteries of IR group and normal control group rats by the whole-cell patch clamp, Western blot and real-time PCR methods. Results   BKCa current density was decreased in smooth muscle cells of aortas and mesenteric arteries in IR group. Compared with that in normal control group, peak BKCa current at +60 mV was significantly decreased after iberiotoxin perfusion at
100 nmol/L (both P<0.01). BKCa β1-subunit expression at the mRNA level and protein level was significant decreased in IR group compared with that in normal control group (P<0.05). Conclusion   Our results suggest that BKCa is decreased both in macrovessels and microvessels in insulin resistance rats. These changes may be related to the down-regulation of β1 subunit expression.
